CJC-1295 / Ipamorelin

A popular combination for raising growth hormone. Each peptide has limited human data, and the combination has none.

Dosing schedules (as studied)

Both schedules come from the 2006 repeat-dose study of CJC-1295 alone in healthy adults. Doses were set per kilogram of body weight, so pick a weight above to see the amount in mg. These were short studies of two or three injections, not long-term regimens.

Every two weeks

Biweekly groups

Two injections, 14 days apart. One group received 30 mcg/kg and another 60 mcg/kg. The 30 mcg/kg dose is shown.

* Limited by the vial's contents or the 28-day use limit after mixing, whichever comes first.

IGF-1 stayed above baseline for up to 28 days after repeat doses.

Supplies for this schedule
  • 1 × 10 mg vials (each vial is used for at most 28 days after mixing)
  • 2 mL bacteriostatic water (2 mL per vial)
  • 2 × 0.5 mL (50-unit) U-100 insulin syringes
  • 5 alcohol prep pads, plus a sharps container

Weekly

Weekly groups

Three injections, 7 days apart. One group received 20 mcg/kg and another 30 mcg/kg. The 30 mcg/kg dose is shown.

Drug levels built up with weekly dosing, peaking 29–70% higher after the third injection than after the first.

Supplies for this schedule
  • 1 × 10 mg vials (each vial is used for at most 28 days after mixing)
  • 2 mL bacteriostatic water (2 mL per vial)
  • 3 × 0.5 mL (50-unit) U-100 insulin syringes
  • 7 alcohol prep pads, plus a sharps container

Reconstitution for this setup

  1. Let the 10 mg vial reach room temperature and wipe both stoppers with alcohol.
  2. Slowly add 2 mL of bacteriostatic water, aiming down the inside wall of the vial.
  3. Swirl gently until clear. Never shake.
  4. Label the vial: 5 mg/mL, with today's date. Refrigerate and use within 28 days.

Study details

StudyDoseSchedule
Study 1 (single dose)30, 60, 125 or 250 mcg/kgOne injection under the skin
Study 2 (repeat doses)30 or 60 mcg/kgTwo injections, 14 days apart
Study 2 (repeat doses)20 or 30 mcg/kgThree injections, 7 days apart
  • The authors reported the drug was best tolerated at 30 or 60 mcg/kg.
  • Growth hormone and IGF-1 stayed raised for days after each dose, with a cumulative effect after repeat doses.
  • Clinical development of CJC-1295 did not continue.

Research summary

CJC-1295 mimics growth-hormone-releasing hormone. Ipamorelin mimics ghrelin. Together they are meant to raise the body's own growth hormone release.

What the research shows

  • A 2006 human study found that CJC-1295 raised growth hormone and IGF-1 levels for days after one dose. Clinical development did not continue.
  • Ipamorelin reached Phase 2 for post-surgical bowel recovery but did not meet its goal.
  • No trials have tested the two together.

Regulatory status

The FDA placed both CJC-1295 and ipamorelin in compounding Category 2 in 2023, citing safety concerns.
